What happens to the PNG's transparency in BMP?

Standard BMP has no usable alpha channel, so transparent regions are flattened onto a solid background color. If the cutout matters, keep a PNG copy — once flattened into BMP, the transparency can't be recovered automatically.

How much will the file size increase?

A lot, and most for the images PNG compresses best. A 200 KB screenshot full of flat color can easily become a 5-6 MB BMP, because BMP writes every pixel raw regardless of how repetitive the image is.

Why would anything still require BMP instead of PNG?

Because a BMP can be read with a few dozen lines of code and no decompression library. Old Windows applications, industrial machines, microcontroller firmware, and some scientific tools were built on that simplicity and never moved on.
